Chromatogram Editing and Assembly in Geneious Pro™
All sequence assembly follows the same steps in Geneious Pro™, whether you're assembling a two strands of chromatogram data in Sanger format or enormous sequence list documents for NGS assembly. Select the sequences you want to assemble (include a reference if you have one) and click assembly to get underway immediately.
Chromatogram Editing and Assembly
- Chromatogram assembly – Assemble chromatograms on their own or to a reference and inspect the the quality of the traces in your chromatograms. Review the assembly by looking at the consensus sequence and the original traces for difficult calls
- Import Sequencher data – Geneious Pro 5.0 can import the common assembly format (.CAF) to rescue your old Sequencher projects.
- Assemble by name – Use this powerful automated workflow to assemble groups of similar sequences into separate contigs based on their name
- Automated trimming – Automatically trim your chromatogram sequences for low quality regions, to remove primer or vector sequences, or any other custom sequence before you assemble
- Disagreements – Jump quickly between disagreements (ctrl+D) in your contig and Geneious Pro will keep a record of all edits you make to your contig in the document history so you always know what's changed
- Chromatogram trace resizing – Resize the peaks of your chromatograms by clicking+holding and moving the mouse up and down to resolve tightly packed peaks
- Call secondary peaks – Download the Heterozygotes plugin to call or annotate the secondary peak information in chromatograms
- Extensive trimming options – Trim off your vector or primer sequence, or any 5' or 3' custom sequence length
