Molecular Cloning in Geneious Pro™

In Geneious Pro it's easy to complete a workflow for in silico cloning.

Restriction Analysis

  • Find restriction sites – Create a list of restriction enzymes and display all of the enzymes on your linear or circular DNA fragment
  • Digest into fragments – Digest a sequence into fragments with a list of restriction enzymes and use the fragments in future analysis
  • Show non-cutting enzymes – Find enzymes that don't cut your sequence(s) or a particular region of your sequence
  • Virtual gel – View your restriction digests on a virtual gel, digesting all at once, or one at a time

in silico Cloning

  • One-step Gateway cloning – Select up to four fragments, four donor vectors and a destination vector, and perform in silico Gateway cloning in a single step.
  • Ligate sequences – Ligate multiple fragments together that have matching overhangs.
  • One-step cloning – Insert a fragment of DNA into a vector with matching overhangs in a single step.
  • TOPO cloning – Select the bases between any two cut-sites and simply cut and paste to perform blunt-end TOPO cloning.
