Geneious Prime 2027.0 Feature Overview

Geneious Prime 2027.0 introduces an Off-Target viewer and CFD scoring for deeper CRISPR off target analysis, with genome searches up to three times faster. It also brings more flexible cloning, including Golden Gate spacers and backbone fragment choice in Gibson and In-Fusion, and adds true circular sequence alignment, plus much more.

Enhanced CRISPR off-target analysis

A new Off-Target viewer surfaces the 100 highest-scoring off-target sites for a guide of interest, so you can investigate each in detail. The CFD off-target score (Doench et al 2016) is now available, and you can run multiple on-target and off-target scoring algorithms in a single operation. Off-target searching across genomes is also up to three times faster, and you can filter results by setting a threshold for each score, returning only guides that meet it.

More flexible cloning

Cloning gives you more control over your backbone and inserts. In Gibson and In-Fusion cloning you can choose any backbone fragment from the tag dropdown when a circular backbone is digested into multiple pieces. Golden Gate cloning now supports adding spacers, and changes to your input documents can be propagated through to a Parts cloning result.

Align circular sequences

You can now align circular sequences. This is specifically for aligning circular sequences: Geneious treats them as circular rather than linear, so you no longer need to linearize them first. Alignments of linear sequences are unaffected.
